About Mesalazine
5-Aminosalicylic Acid Derivative, Anti-inflammatory bowel agent.
Mechanism of Action of Mesalazine
Mesalazine exerts its anti-inflammatory action by inhibiting lipoxygenase enzyme. This will leads to decreased production of pro-inflammatory leukotrienes (LTB4) and 5-hydroxy eicosa tetra enoic acids (HETE) in the intestinal wall. It inhibits the production of inflammatory mediators like interleukin-1, tumor necrosis factor-alpha etc.

Pharmacokinets of Mesalazine
Absorption: Mesalazine is poorly absorbed after oral administration and also after rectal administration.
Distribution: It is uniformly distributed over the inflamed intestinal mucosa. Small amount of the drug is distributed in protein bound form in the systemic circulation.
Metabolism: It is rapidly metabolised in the intestinal mucosal wall in the liver to N-acetyl 5- amino salicylic acid.
Excretion: After oral administration it is excreted in urine and after rectal administration it is excreted in faeces.

1- 3 weeks.
Duration of Action for Mesalazine
12 hours.
Half Life of Mesalazine
5 hours after initial dose.

2.Crohn`s disease
3.Proctitis and Proctosigmoiditis (use only suppositories)
Interactions for Mesalazine

Typical Dosage for Mesalazine
Oral
Adult:
Ulcerative colitis: 400 - 800 mg 3 times daily.
Crohn`s disease: 800 mg 3 times daily.
Suppositories:
Proctitis and Proctosigmoiditis 1 - 2 suppositories are inserted 3 times daily after defecation
Dosage is adjusted depending upon the severity of the disease.
Enema:1 - 4 g is administered and retain for 8 hours.
Children: not recommended

Effects of Missed Dosage of Mesalazine
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.
