About Fluticasone
Synthetic topical glucocorticoid, Anti inflammatory and antipruritic in corticosteroid-responsive dermatoses; atopic dermatitis.
Mechanism of Action of Fluticasone
It reduces prostaglandin synthesis by inhibiting cyclo oxygenase enzymes both COX1 and COX2.In higher doses it reduces the thromboxane A2 synthesis by COX1 inhibition. So platelet aggregation is inhibited by this drug. After topical application in the case of ophthalmic preparation it prevents miosis by reducing ocular prostaglandins synthesis. It is a phenylalkalonic acid derivative Non Steroidal Anti Inflammatory Drug.

Absorption- Well absorbed orally.
Distribution-99% bound to the plasma proteins.
Metabolism- In liver it is metabolized by hydroxylation and conjugation.
Excretion-It is excreted through urine.

1. Rheumatoid arthritis
2. Osteo arthritis
3. Ankylosing spondylitis
4. Pain in patients with dysmenorrhea.
5. Post operative inflammation of the eye.

Typical Dosage for Fluticasone
Oral
Adults- 200-300mg/day in 2-4 divided doses.
Topicaly-0.03% ophthalmic solution. Add 1 drop in every ? hr to the eyes beginning 2-3 hrs before surgery.

Storage Requirements for Fluticasone
Store in a sealed container at room a temperature and avoid moisture.
In the case of ophthalmic preparation stored between 15 and 30 degree Celsius.
Effects of Missed Dosage of Fluticasone
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.
Effects of Overdose of Fluticasone
Supportive and symptomatic treatment should be given. Empty the stomach by emesis or gastric lavage.
