First Generation selective histamine H1 antagonist,Ethanolamine derivative ,anticholinergic, Antihistamine, and sedative.
Mechanism of Action of Clemastine
Clemastine is a selective histamine H1 antagonist. It binds to the histamine H1 receptor present on uterus, GI tract, bronchi and large blood vessels and blocking the action of endogenous histamine( These are responsible for many allergic symptoms) which leads to temporary relief of the negative symptoms caused by histamine.

Metabolism- Metabolized in the liver to its metabolites by mono/didemethylation.
Excretion- Excreted via urine.

CNS depressant: CNS depressant action potentiated.
MAOIs: Prolonged and intensify the anticholinergic effects.
Typical Dosage for Clemastine
Oral-
Rhinitis -
Adult and children (>12yr)-
1.34-2.68mg two times or three times daily.
Children (>12 yrs) - 0.67mg two times daily.
Skin allergies-
Adult and children (>12yr)-
2.68 mg every three times daily.
Children (>12 yrs) -1.34mg every two times daily.

Storage Requirements for Clemastine
Store at room temperature and away from excess heat and moisture. Keep out of reach of children.
Effects of Missed Dosage of Clemastine
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.
Effects of Overdose of Clemastine
Give symptomatic and supportive treatment. Induce emesis and administer charcoal for preventing further absorption of drug. Provide gastric lavage if needed. Treat hypotension with vasopressors and seizures with phenytoin or diazepam.
