Mechanism of Action of Calcitonin
In bone it suppresses resorption of bone by inhibiting the activity of osteoclasts (decrease the ruffled surface which hold the resorptive pit), a cell type that "digests" bone matrix, releasing calcium and phosphorus into blood. In kidney it inhibits tubular reabsorption of the Ca2+ ions, leading to increased rates of their loss in urine.

Intramuscular and subcutaneous- 8 to 24 hrs
Intravenous- ? to 12 hrs

Typical Dosage for Calcitonin
Intramuscular and subcutaneous-
Adult-
Paget`s disease-
Initial dose- 100 IU
Maintenance dose- 50 to 100 IU, three times daily
Hypercalcemia- 4 IU/kg every 12 hrs. The dose increase by 8 IU/kg every 12 hrs
Postmenopausal osteoporosis- 100 IU daily
Intravenous-
Hypercalcemia- 2 to 16 IU/kg every 12 hrs
Intranasal-
Postmenopausal osteoporosis- 200 IU daily

Storage Requirements for Calcitonin
Store in refrigerator, protect from freezing, sealed container.
Effects of Missed Dosage of Calcitonin
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.
Effects of Overdose of Calcitonin
Give symptomatic and supportive treatment. Administer parenteral calcium if there is hypocalcaemia and tetani associated with it occurs
