About Allyloestrenol
Synthetic progestational hormone (Estren derivative progestogen).
Mechanism of Action of Allyloestrenol
It exerts it`s action by stimulating synthesis and secretion of placental progesterone. Progesterone acts through nuclear progesterone receptors situated in female genital tract, breast, CNS, and pituitary. When the drug binds to progesterone receptors it undergoes dimerization and attaches to progesterone response element of target genes and regulates transcription through coactivators; which ultimately leads to modification of protein synthesis. It contributes to proper development and maintenance of female sexual functions. It prepare uterus for nidation and maintenance of pregnancy. It prevents endometrial shedding, decreases uterine motility, and also inhibits immunological rejection of the foetus by decreasing T-cell function and cell mediated immunity.
It also exerts some antiandrogenic effect by selectively taken in to prostate and produce direct competitive antagonism against androgen in the prostate. It selectively inhibits uptake of serum testosterone in to the prostate and also inhibits the reduction of testosterone in to 5-alpha-dihydrotestosterone and formation of 5-alpha-dihydrotestosterone-cytosol receptor complex

Tricyclic Antidepressants, Hypoglycaemic Agents, Anticoagulants: Efficacy of these drugs reduced.
Rifampicin, Barbiturates & Phenytoin: Interfere with the effects of allyloestrenol.

Typical Dosage for Allyloestrenol
Habitual abortion: 5 to 10mg/day when pregnancy is established until one month till critical period is over.
Threatened abortion: 5mg thrice daily for 5 to 7 days then reduces gradually until symptoms disappear.
Threatened premature labour: Dosage based on individual requirements up to 40mg/day
Benign prostatic hypertrophy: 25mg 12th hourly.
