About Nafarelin Acetate
A man-made hormone ,GnRH agonist, Antiendometriotic agent,in Central precocious puberty (CPP).
Mechanism of Action of Nafarelin Acetate
It is a long acting potent gonadotropin releasing hormone (GnRH) agonist. It stimulates the FSH and LH release from the pituitary which results in a temporary increase in the ovarian steroidogenesis. After 3 days of Nafarelin administration pituitary become refractory to further stimulation. Down regulation of pituitary GnRH receptors occurs in 10days which result in inhibition of FSH and LH release which leads to reduced secretion of gonadal steroids. Peak inhibition of gonadotropin release occurs in one month.

Typical Dosage for Nafarelin Acetate
Endometriosis: 200?g twice daily for 6 months taken as one spray to one nostril in the morning and one spray to the other nostril in the evening (400?g/day). Begin treatment between day 2-4 of the menstrual cycle.
Central precocious puberty: 1600?g/day as two sprays (400?g) into each nostril in the morning (4 sprays) and 2 sprays into each nostril in the evening (4 sprays), a total of 8 sprays/day.
