About Norethindrone Acetate
Derivative of 19-nor testosterone, Synthetic Progestogenic hormone, Contraceptive.
Mechanism of Action of Norethindrone Acetate
It is a progestin derivative exerts its action through nuclear progesterone receptors situated in female genital tract, breast, CNS, and pituitary. When the drug binds to progesterone receptors it undergoes dimerisation and attaches to progesterone response element of target genes and regulates transcription through coactivators. It inhibits growth hormone release from pituitary by reinforcement of normal feedback inhibition. It reduces frequency of LH secretary pulses, attenuate LH surge, and suppresses ovulation. It causes thickening of cervical and induces sloughing of the endometrium.

Interactions for Norethindrone Acetate

Typical Dosage for Norethindrone Acetate
Endometriosis: 5mg/day orally for 14 days. Then increases the dosage; gradually to 10mg/day. If bleeding ceases reduce to initial dosage. Continue therapy for 6months.
Dysfunctional uterine bleeding: 5mg 12th hourly for 10 days.
Postponement of menstruation: 5mg 12th hourly 3days before onset of menstruation, but not more than two weeks.
Premenstrual syndrome: 5mg 1 - 2 times daily from 19th to 26th day of menstrual cycle.
Contraception: 0.35mg daily from 1st day of cycle and continue uninterrupted.
Amenorrhea: 2.5 to 10mg for 10 days.
