About Dydrogesterone
It is in the class of progestogens.
Mechanism of Action of Dydrogesterone
It is a progestin derivative exerts it`s action through nuclear progesterone receptors situated in female genital tract, breast, CNS, and pituitary. When the drug binds to progesterone receptors it undergoes dimerization and attaches to progesterone response element of target genes and regulates transcription through coactivators; which ultimately leads to modification of protein synthesis. It contributes to proper development and maintenance of female sexual functions. It prepare uterus for nidation and maintenance of pregnancy. It prevents endometrial shedding, decreases uterine motility, and also inhibits immunological rejection of the foetus by decreasing T-cell function and cell mediated immunity.

Interactions for Dydrogesterone
Cyclosporin: Increased plasma cyclosporin concentration.
Rifamycin: Metabolism accelerated by rifamycins.
Typical Dosage for Dydrogesterone
Dosage:
Oral:
Endometriosis:
5 to 10mg twice daily to thrice daily; continuously or cyclically from 5th day to 21st day of menstrual cycle.
Premenstrual syndrome: 5 to 10mg/day from 12th to 26th day of the menstrual cycle.
Dysmenorrhea: 5 to10mg 12th hourly from 5th day to 25th day of menstrual cycle.
Replacement therapy: 10mg/day for last 14 days of the menstrual cycle. Increases to twice daily if required.
Habitual abortion: 10mg twice daily from 11th to 25th day of menstrual cycle and then continuously after conception during first half of pregnancy and reduces gradually.
Infertility, Irregular cycles: 10mg 12th hourly from 11th to 25th day of menstrual cycle. Repeat as required.
Dysfunctional uterine bleeding: 10mg 12th hourly together with an oestrogen for 5 to 7days.
Prophylaxis: 10mg 12th hourly together with an oestrogen from 11th to 25th day of cycle.

Store in a well closed container at controlled room temperature below 25 degree C. in a dry dark place. Keep out of reach of children.
Effects of Missed Dosage of Dydrogesterone
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.

Give supportive measures and symptomatic treatment.
