Mechanism of Action of Fluconazole (Eye Prep.)
Fluconazole is fungicidal or fungistatic depending on the drug concentrations. It inhibits the conversion of Lanosterol to 14 demethyl Lanosterol by inhibiting the cytochromeP450 enzyme 14 alpha demethylase and impair biosynthesis of ergosterol for the cytoplasmic membrane. This will leads to accumulation of 14 - a-methyl sterols and impair the functions of certain membrane bound enzymes such as ATPase and enzymes of electron transport system and also disrupt the close packing of acyl chains of phospholipids. These will produce inhibition of fungal growth

0.3% Eye drop is available. Initial dose: 1 drop should be instilled in to the affected eye every 4 hours for 3 - 4 days followed by 1 drop every 6 hours for 2 - 3 weeks.
