Mechanism of Action of Tacrolimus Topical
It binds to specific receptors on T cells. This leads to an increase in intracellular calcium that, in turn, causes a series of reactions inhibiting the transcription of several genes, mainly the cytokines (IL-2, IL-4, and IL-5). It also decreases the production of IL-8 receptor genes and inhibits the production of receptors for IgE molecules.

Concomitant administration of known CYP3A4 inhibitors in patients with widespread and/or erythrodermic disease should be done with caution.

For atopic dermatitis-
Adults and Children over 2 to 15 years old -
Gently apply 0.03% or 0.1% ointment to skin that is clean and dry two times a day. Do not cover the area with a bandage that sticks to the skin.
Continue to treat for 1 week after symptoms are gone.
