Mechanism of Action of Ifosfamide
In the liver drug undergo enzymatic activation into 4-hydroxyifosfamide.This alkylates the DNA by binding to N7 position of the guanine residue. This results cross linking/abnormal base pairing/scission of DNA strand. There is cross linking of nucleic acid with proteins can also takes place. The overall process is cell cycle nonspecific reaction.

Absorption- Rapidly absorbed after intravenous administration
Distribution- Widely distributed in the body. It crosses the blood brain barrier.
Metabolism- Metabolized in the liver into its active and inactive metabolites.4-hydroxyifosfamide is its active metabolite.
