About Lomustine
An alkylating nitrosourea compound, Antineoplastic.
Mechanism of Action of Lomustine
This highly lipophilic nitrosourea compound undergoes hydrolysis in vivo to form reactive metabolites. These metabolites cause alkylation and cross linking of DNA. This process slows down the synthesis of cancer cells. This drug also has been shown to affect a number of cellular processes including DNA, protein synthesis, processing of ribosomal &nucleoplasmic messenger RNA, DNA based component structure, the rate of DNA synthesis and DNA polymerase activity. These all processes is cell cycle-phase nonspecific.
Pharmacokinets of Lomustine
Absorption- Rapidly absorbed after oral administration.
Distribution - Widely distributed in the body. It crosses the blood brain barrier.
Metabolism- Metabolized in the liver to its active and inactive metabolites.
Excretion -Primarily excreted via urine. A small portion is excreted through feces and through lungs.

3 hrs after oral administration

16 to 48 hrs
Half Life of Lomustine
Its elimination half life is about 6 hrs for initial phase and 1 to 2 days for terminal phase.

Storage Requirements for Lomustine
Keep in tightly closed container at room temperature, away from heat and moisture. Keep out of reach of children.
Effects of Missed Dosage of Lomustine
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.

Give symptomatic and supportive treatment.
