Natural (Vinca) Alkaloid, antimicrotubule drug, Antineoplastic.
Mechanism of Action of Vinblastine
This antineoplastic alkaloid drug bind to the tubuline.This binding prevents the polymerization of the spindles to microtubules. Thus it inhibits the mitosis at metaphase. These drug also can interfere with the cells ability to synthesize DNA& RNA.Vinblastine also seem to inhibit cancer cells growth, interfering with their glutamic acid metabolism.

Absorption- Rapidly absorbed after intravenous administration.
Distribution- Widely distributed in the body.43-99% bound to the plasma proteins. Drug crosses the blood brain barrier.
Metabolism-Drug metabolized in the liver to its active metabolites Desacetylvinblastine.
Excretion-Excreted via urine and bile.

Interactions for Vinblastine
Mitomycin: Acute shortness of breath and severe bronchospasm.
Phenytoin: Combination chemotherapy (including vinblastine) may reduce plasmalevels and increase seizure activity.
Erythromycin: Severe myalgia, neutropenia and constipation.
Typical Dosage for Vinblastine
Intravenous-
Adult-
0.1-0.5mg/kg every 7 to 10 days or 3.7mg/metre square weekly.
Children-
2.5mg/metre square as a single dose every week.
