Alkylating Agent, Platinum Analog, A systemic antineoplastic Agent
Mechanism of Action of Carboplatin
This platinum co ordination complex hydrolyzed intracellularly to produce a highly reactive moiety, which causes cross linking of DNA at the site of N7 of guanine residue. It can also react with -SH groups in proteins and has radiomimetic property.
Pharmacokinets of Carboplatin
Absorption- Rapidly absorbed after intravenous administration Distribution- Widely distributed to the body water. It does not bind to the plasma proteins. The metal platinum in the drug after degradation bound to the plasma proteins (87%).
Metabolism- Metabolized into metabolites by hydroxylation
Excretion- Excreted through urine. Enterohepatic recirculation is also takes place.

Interactions for Carboplatin
Phenytoin : Serum concentration may be decreased resulting in loss of therapeutic effect.
Typical Dosage for Carboplatin
Intravenous
Adult-
300-600mg/metre square i.v.infusion every 3-4 weeks
For brain tumors in children-
175mg/metre square weekly for 4 weeks, with a 2 weeks recovery period between courses

Store at 15 -30 degree C temperature. Protect from light.
Effects of Missed Dosage of Carboplatin
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.
