Serotonin 5-HT1B, 1D Receptor Agonist, Antimigraine.
Mechanism of Action of Rizatriptan
Rizatriptan is a 5-hydroxytriptamine1B/1D receptor agonist. Rizatriptan relieves migraines by constricting cranial blood vessels of the carotid circulation and reduces blood flow. It also inhibits the release of neuropeptides responsible for pain.

Absorption: Rizatriptan is absorbed after oral administration and its bioavailability is about 45%
Distribution: It is distributed minimally in protein bound form.
Excretion: Rizatriptan undergoes metabolism via oxidative deamination. It is excreted primarily in urine and small amount in faeces.

Interactions for Rizatriptan
Antibacterials: quinolones possibly reduce metabolism of Rizaitriptan. Avoid concomitant use with clarithromycin and erythromycin.
Antidepressants: risk of CNS toxicity with MAOIs.
Beta blockers: Propranolol may increase plasma concentration of Rizatriptan.
Ergotamine: Increased risk of vasospasm.

Typical Dosage for Rizatriptan
Adult: 5 - 10mg at the beginning of headache and if sufficient response does not obtain then repeat the dose after 2 hours, maximum dose: 30mg / day. Patient taking Propranolol the maximum dose is 15mg/ day
Children: Not recommended

Effects of Overdose of Rizatriptan
Give supportive measures and symptomatic treatment. Drug can be removed from the body by gastric lavage. Absorption of the drug can be reduced by administration of activated charcoal.
