Third Generation Cephalosporin Parenteral antibiotic.
Mechanism of Action of Ceftizoxime
Ceftizoxime is a 3rd generation cephalosporin which exerts its bactericidal action against both gram positive & gram negative organisms by inhibiting bacterial cell wall synthesis. Ceftizoxime inhibits transpeptidase & thus prevents cross linking of bacterial cell wall. Transpeptidase & associated proteins constitute various types of specific binding proteins which have affinity for cephalosporins like Ceftizoxime
Pharmacokinets of Ceftizoxime
Absorption: Orally not well absorbed & should be given parenterally, Distribution: widely distributed, & good CSF penetration, Metabolism: Not metabolized inside the body, Excretion: Excreted mainly through urine & small amount is seen in breast milk.

1.Meningitis
2.Resistant hospital acquired infections
3.Septicemia
4.Infections in immunocompromised patients
5.Urinary tract & genital tract infections

8.Bone & Joint infections
9.Gonorrhea.
Interactions for Ceftizoxime
Aminoglycosides, other cephalosporins and frusemide : Nephrotoxicity potentiated.
Oral anticoagulants : Action potentiated

Typical Dosage for Ceftizoxime
Adults: 1g to 2gm I.M. or I.V. Once daily or two times daily
Maximum adult dose: Up to 4gm/day
Meningitis & Endocarditis:1 to 2gm I.M. or I.V twice daily; two weeks for meningitis & four weeks for endocarditis
Gonorrhea: 250mg once daily

Children: 50 to 100 mg/kg/day I.M or I.V. two times daily
Maximum child dose: 2g/day
Meningitis & Endocarditis: 50 to 100 mg/kg/day twice daily; two weeks for meningitis & four weeks for endocarditis

Effects of Missed Dosage of Ceftizoxime
Take the missed dose as soon as noticed and if it is the time to take the next dose then skip the missed dose.
Effects of Overdose of Ceftizoxime
Symptoms of over dosage include neuromuscular disorders & some times seizure may get precipitated. Treatment includes removal of drug from the body by dialysis & supportive measures
