Mechanism of Action of Prothionamide
Prothionamide is bactericidal or bacteriostatic depending on the concentration of the drug at the site of infection and the susceptibility of the organism. It is an Ethionamide derivative. Pothionamide inhibits mycolic acid synthesis and leads to inhibition of cell wall formation.
Pharmacokinets of Prothionamide
Absorption: It is well absorbed after oral administration. Distribution: It is widely distributed in the body. Metabolism: It undergoes metabolism in the liver to produce active sulphoxide metabolite and inactive metabolite. Excretion: It is excreted mainly through urine.

Oral hypoglycaemic agents: Alters the actions of OHAs and disturbs blood glucose levels.
INH: Decreases serum-INH levels.
Typical Dosage for Prothionamide
Adult: 250 - 500 mg two times daily.
Leprosy: 15 - 20 mg / kg body weight / day; Maximum daily dose 1g
Children: 10 - 20 mg / kg body weight / day in two divided dose.
Leprosy: 12 - 15 mg / kg body weight / day; Maximum daily dose 750 mg
