Dihydrofolate reductase inhibitor,biguanide, A prophylactic Antimalarial.
Mechanism of Action of Proguanil
In vivo Proguanil undergoes chemical modification and form Cycloguanil.This Cycloguanil has high affinity to bind with plasmodial dihydrofolate reductase enzyme and inhibits the reduction of Dihydrofolic acid (DHFA) to Tetra hydrofolic acid (THFA) .This will interferes the synthesis of folic acid in malarial parasite.
