About Ganciclovir
A synthetic acyclic purine nucleoside analog of 2`-deoxyguanosine, Antiviral.
Mechanism of Action of Ganciclovir
Ganciclovir is converted to Ganciclovir triphosphate by Kinases enzyme in the virus. This Ganciclovir triphosphate competitively inhibits DNA polymerase enzyme and inhibits DNA synthesis in the virus. Ganciclovir triphosphate is also incorporated in viral DNA and stops the DNA elongation and permanent inactivation.
Pharmacokinets of Ganciclovir
Absorption: It is poorly absorbed after oral administration. Distribution: It preferentially concentrates in Cytomegalovirus. Metabolism: Ganciclovir is converted in to Ganciclovir triphosphate by kinase enzyme in the virus. Excretion: It is excreted mainly in the urine.

Typical Dosage for Ganciclovir
Oral:
CMV prophylaxis: 1g to be taken every 8 hours.
IV
Cytomegalovirus retinitis: 5mg/kg two times daily for 2 - 3 weeks. Maintenance dose: 5mg/kg once daily for 1 week

Store at room temperature between 15-30 degrees C. Keep out of the reach of children.
Effects of Missed Dosage of Ganciclovir
Take the missed dose as soon as remember and if it is almost time for the next dose, skip the missed dose and continue regular schedule. Do not take a double dose.
Effects of Overdose of Ganciclovir
Give supportive measures and symptomatic treatment. Haemodialysis can be done if needed.
