Norepinephrine and dopamine reuptake inhibitor,nicotinic antagonist, aminoketone derivative, Atypical antidepressant, Smoking Cessation Aid.
Mechanism of Action of Bupropion
Bupropion blocks the re-uptake of neurotransmitter Dopamine and Noradrenaline from the synapse in to the presynaptic nerve terminal in the CNS. It has only a less activity in blocking the reuptake of Serotonin. Bupropion potentiates the noradrenergic and Dopaminergic activity and produces antidepressant activity.
Smoking cessation action: Bupropion is a competitive antagonist of nicotine.

Absorption: Bupropion is well absorbed after oral administration but its bioavailability is 5 - 20 % in animals. Distribution: It is distributed in the body in protein bound form. Metabolism: It undergoes metabolism in the liver and produces active metabolites Excretion: Excreted primarily in urine and rest in faeces.

Typical Dosage for Bupropion
Adult
Depression:
Initial dose: 150 - 200 mg / day in 2divided doses. If sufficient response do not occur after 3 days dose can be increased to 300 mg / day in 3 divided doses.
Maximum dose: 450 mg / day
Smoking cessation treatment: Initial dose: 150 mg daily for 3 days then dose is increased to 300 mg / day in 2 divided doses for 7 - 12 weeks

Store at 15 - 30 degree C in a tightly closed container. Protect from direct light and heat
Effects of Missed Dosage of Bupropion
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.
Effects of Overdose of Bupropion
Give supportive measures and symptomatic treatment. Drug can be removed from the body by inducing emesis and by gastric lavage. Activated charcoal can be given to reduce the absorption of the drug. Seizure can be treated with IV Benzodiazepines
