About Mephentermine
Alpha-adrenergic agonist, Phenethylamine derivative, A cardiac stimulant.
Mechanism of Action of Mephentermine
It exerts it`s pharmacological action by directly activating alpha and beta adrenergic receptors and by releasing nor adrenaline. It produces both cardiac stimulation and vasoconstriction. It increases cardiac out put and systolic and diastolic BP. The direct positive chronotropic effect is counterbalanced by vagal stimulation due to rise in mean BP.

Absorption: Absorbed orally and also administered parenterally, Metabolism: Metabolized mainly by demethylation and also by hydroxylation. Excretion: Excreted through urine both as unchanged drug and metabolites.

1. Maintenance of BP in hypotensive states
2. Shock in myocardial infarction.
Interactions for Mephentermine
Cyclopropane & Halothane: May result in serious ventricular arrhythmias.
Digitalis: Increased risk of cardiac arrhythmias.
Phenothiazines, TCAs: May antagonise pressor response of mephentermine.
MAOIs: Potentiate pressor effects of mephentermine.
Oxytocic drugs: May cause severe persistent hypertension.
Typical Dosage for Mephentermine
Oral: 20mg once daily or 12th hourly.
Injection: 15 to 60mg as required by the patient based on response.
