A xanthine derivative, Phosphodiesterase inhibitor, Blood viscosity reducing agent.
Mechanism of Action of Pentoxifylline
The drug exerts its pharmacological action by improving rheological (flow) properties of blood. It improves blood flow to ischaemic areas by increasing capillary blood flow by improving erythrocyte flexibility and reducing blood viscosity. It inhibits phosphodiesterase enzyme.
Pharmacokinets of Pentoxifylline
Absorption: Well absorbed orally but bioavailability decreases due to first pass metabolism. Distribution: Distributed in to erythrocyte membrane. Metabolism: Extensively metabolized in erythrocytes and liver. Excretion: Excreted mainly through urine and a small portion is excreted through faeces.

1.Intermittent claudication
2.Occlusive arterial diseases of limbs
3.Cerebrovascular insufficiency
4.Circulatory disorder due to arteriosclerosis

7.Other chronic occlusive vascular disorders
Interactions for Pentoxifylline
Warfarin: Bleeding and prolonged prothrombin time in patients receiving pentoxifylline with or without anticoagulants or platelet aggregation inhibitors. PT(Prothrombin Time) should be monitored in patients on warfarin.
Typical Dosage for Pentoxifylline
Oral: 400mg 12th hourly to 8th hourly
I.V.: 300mg/15ml in 300ml normal saline slowly infused over 3 hours; 12th hourly to 8th hourly.

Store in a dry place at a temperature range of 15 to 30 degree C. Protect from light
Effects of Missed Dosage of Pentoxifylline
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.
Effects of Overdose of Pentoxifylline
Treatment is supportive and symptomatic. Empty stomach by gastric lavage and reduce absorption by administration of activated charcoal. Monitor and assist respiratory function and blood pressure.
