Reversible Monoamine oxidase (MAO-A) inhibitors, Antidepressant.
Mechanism of Action of Moclobemide
Moclobemide is a reversible inhibitor of Monoamine oxidase enzyme. It selectively inhibits MAO-A than MAO-B.It decreases deamination of Serotonin, Noradrenaline and Dopamine. This will result an increase in concentration of neurotransmitters and produces antidepressant activity

Absorption: It is well absorbed after oral administration. Distribution: Only 50 % of the drug is bound to plasma protein and is extensively distributed to tissues. Metabolism: It undergoes metabolism in the liver and form inactive metabolites. Excretion: Drug and metabolites are excreted through urine.

Indications for Moclobemide
Depression
Interactions for Moclobemide
Analgesics: CNS excitation or depression (hypertension or hypotension) with codeine, pethidine and possibly fentanyl, morphine and other opioid analgesics. Effects of ibuprofen and possibly other NSAIDs enhanced.
Other antidepressants: Moclobemide should not be started until at least a week after a tricyclic or related antidepressant or an SSRI or related antidepressant has been stopped (2 weeks in the case of paroxetine and sertraline, and at least 5 weeks in the case of fluoxetine), or at least a week after an MAOI has been stopped.
Dopaminergics: Hypertensive crisis with levodopa.
5HT 1 agonist: Risk of CNS toxicity (reduce dose of zolmitriptan).
Sympathomimetics: Avoid sympathomimetics like ephedrine, psuedoephedrine, and phenylpropanolamine.
Ulcer healing drugs: Cimetidine inhibits metabolism ( increases plasma-moclobemide concentration).
Tyramine rich foods: Moclobemide is likely to cause less potentiation of the presor effect of tyramine than the traditional MAOIs, but patients should be advised to avoid consuming large amounts of tyramine-rich foods (like mature cheese, yeast extracts and fermented soya bean products).
Typical Dosage for Moclobemide
Initial dose: 300 mg / day in 3 divided doses.
Dose can be gradually increases to 600 mg / day according to therapeutic response.

Store at 15 - 30 degree C. Keep out of the reach of children.
Effects of Missed Dosage of Moclobemide
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose.
Effects of Overdose of Moclobemide
Give supportive measures and symptomatic treatment. Drug can be removed from the body by inducing emesis and by gastric lavage. Charcoal can be given to reduce the absorption of the drug.
