Potent D2antagonist,butyrophenone derivative, First-Generation Antipsychotic, Typical Antipsychotic,Antiemetic.
Mechanism of Action of Droperidol
Droperidol acts at subcortical level and block the receptors. Droperidol is a potent Dopamine receptor (D2) antagonist.
It also possesses some antihistaminic and antiserotonin activity. It act centrally and produce Antiemetic action.
Pharmacokinets of Droperidol
Absorption: It is well absorbed after IM/IV administration. Distribution: Drug crosses placenta and penetrates in to brain. Metabolism: Drug is metabolised in the liver. Excretion: Drug and metabolites are excreted in urine and faeces.

1.Preanaesthetic medication
2.General anaesthesia (as an adjunct for induction and maintenance of anaesthesia)
3.Adjunct to regional anaesthesia

5.Neuroleptanalgesia
Interactions for Droperidol
Alcohol & other CNS depressants: Additive CNS depression.
TCAs & Antiparkinsonism drugs: Anticholinergic effects potentiated.
Clonidine & Methyldopa: Hypotensive effect of these drugs inhibited.
MAOIs: Orthostatic hypotension with MAOIs.
Typical Dosage for Droperidol
Adults :( IM / IV injection)
Pre-anaesthetic medication during diagnostic procedure as (IM): 2.5 - 10 mg 30 - 60minutes before induction of general anaesthesia
Adjunct to regional anaesthesia: 2.5 - 5 mg as IM or slow IV injection
Antiemetic in cancer chemotherapy: 6.25 mg as IM or slow IV injection
General anaesthesia (as an adjunct for induction and maintenance of anaesthesia): 1.25 - 2.5 mg IV
Neuroleptanalgesia: 2.5 mg with Fentanyl 50 mcg as IV
Children: 0.88 - 0.165 mg / kg body weight IV / IM
