First-Generation Antipsychotic, a thioxanthene derivative, Typical Antipsychotic.
Mechanism of Action of Flupenthixol
Flupenthixol exerts its antipsychotic action by blocking post synaptic Dopamine receptor in the limbic system. It is a potent antagonist of D1&D2 receptors.
Flupenthixol is an alpha adrenergic receptor blocker and depresses many hypophyseal and hypothalamic hormones. It also blocks Prolactin inhibitory factor (PIF) and results in hyperprolactinemia.

Absorption: Flupenthixol is slowly and incompletely absorbed after oral administration. Its bioavailability is only 50 % because it undergoes first pass metabolism. Distribution: It is distributed widely in the body in protein bound form. Metabolism: It is metabolised in the liver to inactive metabolites. Excretion: It is excreted mainly through faeces and small amount through urine.

Typical Dosage for Flupenthixol
Adult:
Depression: 0.5 mg 2 times a day if needed dose can be increased to 2 mg / day after 1 week.
Maximum dose: 3 mg / day
Psychoses: 3 mg 2 times daily dose can be gradually increase to 18 mg / day if needed.
DEPOT INJECTION:
Test dose: 20 mg after 1 week 20 - 40mg and repeat it every 2 - 4 week. Adjust according to response.
Maximum dose: 400 mg /week
