Selective Serotonin 5-HT1B, 1D Agonist, Antimigraine Agent.
Mechanism of Action of Sumatriptan
Sumatriptan is a selective Serotonin (5HT1D) receptor agonist. It selectively constricts the large intracranial blood vessels of the carotid circulation and reduces the blood flow and relieves the migraine. Sumatriptan is used for the acute treatment of migraine attacks and of cluster headache.

Pharmacokinets of Sumatriptan
Absorption: Sumatriptan is very poorly absorbed after oral administration due to first pass effect. It is very rapidly absorbed after subcutaneous administration.Distribution: It is mainly distributed in unbound form.
Metabolism: 80% of the drugs undergoes metabolism in the liver and form inactive metabolites.
Excretion: Excrete mainly in the urine.

7. Hemiplegic migraine
8. Patient on MAO inhibitor or ergotamine therapy

Indications for Sumatriptan
1. Acute attack of migraine treatment with or without aura
Interactions for Sumatriptan
Ergotamine containing preparations: Concomitant use should be avoided because of vasospastic reactions.
MAOIs: Concomitant use within 2 weeks of discontinuation of MAOIs is contraindicated.
Lithium: Concomitant use must be avoided.

Typical Dosage for Sumatriptan
Adult:
Oral: 50 - 100 mg given as soon as symptoms begin repeat the dose if needed after 2 hours. Maximum dose: 300 mg / day.
Parenteral (Subcutaneous): 6 mg as a single dose repeat the dose if needed after 1 hour up to 12 mg/ day.
Children: Not recommended
