About Ergometrine
Ergot alkaloid,an ergoline (and lysergamide) derivative, Sympatholytic, Oxytocic.
Mechanism of Action of Ergometrine
It act on two types of receptors (5HT and alpha receptors) and stimulate them. These receptors are found in the walls of blood vessels and in the uterus (womb). When the receptors are stimulated they cause constriction of the blood vessels and contraction of the uterus. Intense contraction of the muscles of the uterus causes the blood supply of the placenta (placental bed) to shrink and it prevents the bleeding.

Intramuscular- 2-5 minutes

Intravenous- 45 minutes
Intramuscular- 3 hours or longer

Contra-indications of Ergometrine
1. Hypersensitivity to the drug
2. Abdominal pain and bleeding during pregnancy
3, Blood poisoning (toxemia) in pregnancy
4. Decreased kidney function
5. Decreased liver function
6. Decreased lung function
7. Heart disease
8. High blood pressure
9. Induction of labour
10. Infection of the blood or body tissues with pus-forming or other pathogenic organisms (sepsis)

1. Calcium deficiency

3. More than one baby in the uterus (multiple pregnancy)

Typical Dosage for Ergometrine
Prophylaxis of postpartum haemorrhage and postabortion haemorrhage
200 micrograms intramuscularly
200 micrograms may be injected intravenously. IV doses should be given slowly, over a period of at least 1 minute.
Treatment of postpartum haemorrhage and postabortion haemorrhage
200 micrograms may be injected intramuscularly.

The ampoules are to be stored at 2-8?C and protected from light.
Effects of Missed Dosage of Ergometrine
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.
