About Erythropoietin
A glycoprotein hormone produced by kidney, Colony Stimulating Factor, Erythropoiesis-stimulating agents, antianemic agent.
Mechanism of Action of Erythropoietin
It induces erythropoiesis in a dose dependent manner by binding to specific receptors on target cell surface and alters phosphorylation of intracellular proteins. It acts on erythroid marrow stimulates proliferation of colony forming cells of the erythroid series. It induces haemoglobin formation, erythroblast maturation and releases reticulocytes in circulation.

Side Effects of Erythropoietin
1. Exacerbate hypertension

Contra-indications of Erythropoietin
1. Hypersensitivity to the drug or mammalian cell products or albumin
2. In patients with uncontrolled hypertension
Special Precautions while taking Erythropoietin
1.Haematocrit should be determined once each week to measure the initial response.
2.Blood pressure must be monitored closely throughout treatment to help titrate the dose.
3.For optimal response to therapy adequate stores of iron are required. The response to therapy is blunted in patients of iron defeciency. If hypertensive encephalopathy occurs, treatement is stopped.

Typical Dosage for Erythropoietin
Dosage:
I.V.: Starts with 25 to 50i.u./kg over 1 to 2 minutes; thrice weekly. Increases the dosage in increments of 25i.u./kg at intervals of four weeks based on patient`s response.
Maintenance dosage is individualized; usually Maintenance dosage weekly is; 100 to 300i.u./kg in two to three injections.

Store at a temperature range of 15 to 30 degree C.
Effects of Missed Dosage of Erythropoietin
